Analysis of the Phenol and Flavonoid Content from Basil Leaves (Ocimum Americanum L) Extract Using Pulsed Electric Field (PEF) Pre-Treatment CIGR Journal
Hendrawan, Yusuf; Sabrinauly, Sabrinauly; Hawa, La Choviya; Argo, Bambang Dwi; Rachmawati, Muchnuria.
Basil leaves (Ocimum Americanum Linn) are commonly known to contain phenol and flavonoid compounds which can act as antimicrobial and antioxidants. In general, the bioactive compound can be obtained by conventional extraction method, maceration. To increase the amount of bioactive compounds which can be extracted, it is necessary to conduct pre-treatment using Pulsed Electric Field (PEF). PEF has an advantage in increasing the compound transfer contained in cells to be extracted from the plant tissues via electroporation mechanisms, without causing significant temperature changes. This study aims to determine the effect of electric field strength variations (2, 3, and 4 kV/cm) and duration of pre-treatment (1, 2, and 3 minutes) to total phenol and...

Antioxidants properties of chocolates sold in Peru Acta Agron. (Palmira)
Calixto-Cotos,María Rosario; Chire-Fajardo,Gabriela Cristina; Orihuela-Rivera,Carmen Adela.
Abstract The goals of this paper were to evaluate total polyphenol content (TPC), total flavonoid content (TFC), anti-oxidant capacity (AC) and reducing power (RP) of eight dark chocolates that are sold in Peru. Imported and domestic chocolate samples containing between 50% and 74% cacao were defatted. Defatted chocolate (DCh) was separated and treated to extract phenolic compounds. The TPC and AC were determined by using the Folin-Ciocalteu reagent and two in vitro models based on the free-radical capturing capacity DPPH and ABTS. The RP was evaluated by using the potassium pherricyanide method; in the TFCs assessments, catechin was used as the standard. Chocolate TPC varied between 1.69 ± 0.02 and 5.39 ± 0.17 mg gallic acid/g chocolate and AC (DPPH-IC50)...

Apigenin inhibits cell migration through MAPK pathways in human bladder smooth muscle cells Biocell
Qingxin,Liu; Xianggui,Chen; Guolin,Yang; Xuewen,Min; Maoxian,Deng.
Apigenin, a nonmutagenic flavonoid, has been shown to possess free radical scavenging activities, anticarcinogenic properties, antioxidant and anti-inflammatory effects. Recently, apigenin was reported to cause gastric relaxation in murine. To assess possible effects of apigenin on migration of bladder smooth muscle (SM) cell, we isolated SM cells from peri-cancer tissue of human bladder and established a cell model that was capable to overexpress transiently MEKK1 (MEK kinase 1). Results showed that overexpression of active human MEKK1 by adenoviruses infection induced migration of human bladder smooth muscle (hBSM) cells and phosphorylation of MAPKs, ERK, JNK and p38, which are the downstream molecules of MEKK1. Then, hBSM cell overexpressing MEKK1 were...

SOARES,JULIANA M.; FARIA,BRUNA M. DE; ASCARI,LUCAS M.; ALVES-LEON,SONIZA V.; SOUZA,JORGE M. DE; SOARES,ANTONIO G.; CORDEIRO,YRAIMA; ROMÃO,LUCIANA F..
Abstract: Diosmin is a flavone glycoside clinically used as the main component of Daflon for the treatment of venous diseases. Several studies demonstrated that this natural compound can induce apoptosis in different tumors. However, isolated diosmin has not been studied regarding its effects on glioblastoma so far. Since glioblastoma is a highly lethal and fast-growing brain tumor, new therapeutic strategies are urgently needed. Herein, we evaluated the role of this flavonoid against glioblastoma cells using in vitro assays. Diosmin significantly reduced the viability of GBM95, GBM02, and U87MG glioblastoma cells, but not of healthy human astrocytes, as verified by MTT assay. Vimentin immunostaining showed that diosmin induced morphological changes in...

Do different casein concentrations increase the adverse effect of rutin on the biology of Anticarsia gemmatalis Hübner (Lepidoptera: Noctuidae)? Neotropical Entomology
Salvador,Mariana C; Boiça Jr,Arlindo L; Oliveira,Maria C N De; Graça,José P Da; Silva,Débora M Da; Hoffmann-Campo,Clara B.
The flavonoid rutin is recognized as playing an important role in the protection of plants against lepidopterans. Bioassays with this compound are generally carried out using artificial diets. Proteins of high energy value, such as casein, are important ingredients of insect artificial diets as a source of essential amino acids. However, such proteins can generally increase the allelochemical activity. Our objective was to evaluate the effects of rutin on larvae of the velvetbean caterpillar Anticarsia gemmatalis Hübner by incorporating this allelochemical into diets with different concentrations of casein. Three casein concentrations (0, 7 g, or 14 g) combined with none, 0.65%, or 1.30% of rutin were added to the rearing diet and offered to the larvae...

Abstract The response of plants to grazing includes the production of chemical defense compounds such as proteases inhibitors and secondary metabolites as flavonoids, which makes them less palatable to feeding and negatively affecting the physiology of insects. The aim of this study was to evaluate the phytochemical response of soybean cultivars (Glycine max (L.) Merrill) resistant (IAC-17, IAC-24) and susceptible (IAC-P1) to insects after mechanical damage. These cultivars were mechanically injured, and after 24 hours samples of these plants were analyzed by HPLC to identify and quantify flavonoids. The flavonoids daidzein, quercetin, and rutin were quantified, with the highest concentration of daidzin in soybean cultivars after mechanical damage. Rutin...

Plant Polyphenol Antioxidants and Oxidative Stress Biol. Res.
URQUIAGA,INES; LEIGHTON,FEDERICO.
In recent years there has been a remarkable increment in scientific articles dealing with oxidative stress. Several reasons justify this trend: knowledge about reactive oxygen and nitrogen species metabolism; definition of markers for oxidative damage; evidence linking chronic diseases and oxidative stress; identification of flavonoids and other dietary polyphenol antioxidants present in plant foods as bioactive molecules; and data supporting the idea that health benefits associated with fruits, vegetables and red wine in the diet are probably linked to the polyphenol antioxidants they contain. In this review we examine some of the evidence linking chronic diseases and oxidative stress, the distribution and basic structure of plant polyphenol antioxidants,...
